Abstract
Chronic exposure to low concentration arsenic species in food and water poses a health risk. Complex process is involved in absorption of arsenic through intestinal epithelial cells. Herein, IEC-6 (rat intestinal epithelial cell line) cells has been selected to evaluate the toxicological effect of NaAsO2(As III) and Na3AsO4(As V) on growth and function of intestinal epithelial cells. Half maximal inhibition concentration (IC50) of NaAsO2(12mM) and Na3AsO4(48 mM) of 24 hr has been obtained from MTT assay. Cytotoxicity and genotoxicity has beenevaluated by exposure of IEC-6 cells to arsenate (5mM)and arsenite (1.5mM)at low concentrations. The current study demonstrates that trivalent (As III) oxidation state induces ROS, genotoxicity and alters mitochondrial membrane potential (Øm). Whereas pentavalent (AsV) oxidation state induces cytotoxicity and fails to generate genotoxicity at low concentrations. (i.e., 10% of IC50 value) in IEC-6 cells.
